The finding follows closely on  research demonstrating how the drug worked against the Middle East  Respiratory Syndrome (MERS) virus, a related coronavirus. <\/p>\n\n\n\n<figure class=\"wp-block-image size-large\"><img loading=\"lazy\" decoding=\"async\" width=\"580\" height=\"326\" src=\"https:\/\/www.potentialloops.org\/wp-content\/uploads\/2020\/04\/2AP1TD2-b598c7937e0cb7c3ddb3d98f6d897d82.jpg\" alt=\"\" class=\"wp-image-723\" srcset=\"https:\/\/www.potentialloops.org\/wp-content\/uploads\/2020\/04\/2AP1TD2-b598c7937e0cb7c3ddb3d98f6d897d82.jpg 580w, https:\/\/www.potentialloops.org\/wp-content\/uploads\/2020\/04\/2AP1TD2-b598c7937e0cb7c3ddb3d98f6d897d82-300x169.jpg 300w, https:\/\/www.potentialloops.org\/wp-content\/uploads\/2020\/04\/2AP1TD2-b598c7937e0cb7c3ddb3d98f6d897d82-360x202.jpg 360w\" sizes=\"auto, (max-width: 580px) 100vw, 580px\" \/><\/figure>\n\n\n\n<h3 class=\"wp-block-heading\"> Scientists at the University of Alberta have shown that the drug  remdesivir is highly effective in stopping the replication mechanism of  the coronavirus that causes COVID-19, according to new research  published today in the <em>Journal of Biological Chemistry<\/em>. <\/h3>\n\n\n\n<p>The paper follows closely on research published by the same lab in \nlate February that demonstrated how the drug worked against the Middle \nEast Respiratory Syndrome (MERS) virus, a related coronavirus.<\/p>\n\n\n\n<p>&#8220;We were optimistic that we would see the same results against the \nSARS-CoV-2 virus,&#8221; said Matthias G\u00f6tte, chair of medical microbiology \nand immunology at U of A.<\/p>\n\n\n\n<p>&#8220;We obtained almost identical results as we reported previously with \nMERS, so we see that remdesivir is a very potent inhibitor for \ncoronavirus polymerases.&#8221;<\/p>\n\n\n\n<p>G\u00f6tte&#8217;s new paper demonstrates how remdesivir, developed in 2014 to \nfight the Ebola epidemic, works in detail. He likens the polymerase to \nthe engine of the virus, responsible for synthesizing the virus&#8217; genome.<\/p>\n\n\n\n<p>&#8220;If you target the polymerase, the virus cannot spread, so it&#8217;s a very logical target for treatment,&#8221; G\u00f6tte said.<\/p>\n\n\n\n<p>The lab&#8217;s work shows how remdesivir tricks the virus by mimicking its building blocks.<\/p>\n\n\n\n<p>&#8220;These coronavirus polymerases are sloppy and they get fooled, so the\n inhibitor gets incorporated many times and the virus can no longer \nreplicate,&#8221; G\u00f6tte explained.<\/p>\n\n\n\n<p>He said the evidence from his group, along with previously published \nstudies in animal and cell culture models, means that remdesivir can be \nclassified as a &#8220;direct-acting antiviral&#8221; against SARS-CoV-2, a term \nfirst used to describe newer classes of antivirals that interfere with \nspecific steps of the hepatitis C virus (HCV) life cycle.<\/p>\n\n\n\n<p>He said the discovery of that direct action reinforces the promise of\n clinical trials for remdesivir in COVID-19 patients, which are already \nunderway around the world.<\/p>\n\n\n\n<p>While G\u00f6tte said the evidence justifies clinical trials, he cautioned\n that the results obtained in the lab cannot be used to predict how the \ndrug will work with people.<\/p>\n\n\n\n<p>&#8220;We&#8217;ve got to be patient and wait for the results of the randomized \nclinical trials,&#8221; said G\u00f6tte, whose research was funded by the Canadian \nInstitutes of Health Research, Alberta&#8217;s Major Innovation Fund and \nGilead Sciences, which manufactures remdesivir.<\/p>\n\n\n\n<p>The G\u00f6tte lab previously worked on human immunodeficiency virus (HIV)\n and HCV, but a couple of years ago switched to focus on viruses with \nthe highest epidemic potential. The World Health Organization (WHO) \nissued its list of the top pathogens likely to cause severe outbreaks, \nincluding Ebola, Lassa and coronaviruses, in 2015.<\/p>\n\n\n\n<p>&#8220;In that sense we were prepared because my lab specializes in viral \npolymerases,&#8221; said G\u00f6tte, adding that his next step will be to use his \nlab&#8217;s tools to evaluate other promising antivirals.<\/p>\n\n\n\n<p>He is optimistic that the unprecedented amount of research going on \nworldwide and the high level of co-operation between researchers will \nlead to the discovery of one or more effective treatments for COVID-19.<\/p>\n\n\n\n<p>&#8220;We are desperate, but we still have to keep the bar high for anything that we put into clinical trials,&#8221; he said.<\/p>\n\n\n\n<p>Remdesivir is one of several drugs being fast-tracked into trials by \nthe World Health Organization, comparing potential treatments in \nhospitalized COVID-19 patients in a dozen countries, including Canada. \nG\u00f6tte said we can expect results from important clinical trials as early\n as April or May.<\/p>\n\n\n\n<p>G\u00f6tte said it is disappointing that antivirals discovered at the time\n of the severe acute respiratory syndrome (SARS) outbreak of 2003 &#8212; \nwhich might have been effective against COVID-19 too &#8212; were never \ntranslated into widely available treatments, largely because of the huge\n cost involved in developing new drugs.<\/p>\n\n\n\n<p>&#8220;This time around it&#8217;s obvious that we have to cross the finish line,&#8221; he said.<\/p>\n\n\n\n<p>&#8220;Ten billion dollars, it seems a lot, a huge amount,&#8221; G\u00f6tte said.  &#8220;But in the context of this pandemic and the costs associated with this  pandemic, it&#8217;s nothing.&#8221;<\/p>\n\n\n\n<p><strong>Story Source:<\/strong><\/p>\n\n\n\n<p><a rel=\"noreferrer noopener\" href=\"https:\/\/www.folio.ca\/u-of-a-virology-lab-finds-drug-originally-meant-for-ebola-is-effective-against-a-key-enzyme-of-coronavirus-that-causes-covid-19\/\" target=\"_blank\">Materials<\/a> provided by <a rel=\"noreferrer noopener\" href=\"https:\/\/www.ualberta.ca\/medicine\" target=\"_blank\"><strong>University of Alberta Faculty of Medicine &amp; Dentistry<\/strong><\/a>.
